Development of Assembloid-Based Organogenesis Models for Liver Fibrosis Recovery and Disease ModelingNatesh Parashurama
• Led the development of assembloid-based models for organogenesis and liver fibrosis research using iPSCs, advancing tissue regeneration and disease modeling techniques. • Conducted cell culture and differentiation experiments, creating and merging spheroids from HEPG2-WT, fibroblasts (MRC5, HFF), and iPSCs to form Assembloids, while optimizing protocols for tissue engineering applications. • Performed qPCR, immunofluorescence, cellular viability assays, and ImageJ analysis to assess the impact of drug inhibition on liver development, focusing on critical signaling pathways in liver organogenesis. • Trained and supervised over 10 team members in advanced molecular biology techniques, ensuring lab safety compliance and promoting experimental rigor in BSL-2 laboratory environments. • Published key findings on in vitro liver organogenesis and Assembloids formation, significantly contributing to the field of synthetic tissue engineering and liver fibrosis treatment research.

Comparative Decellularization Protocols for Extracellular Matrix Isolation and Characterization in Pulmonary Fibrosis ResearchRuozang Zhao
• Focused on pulmonary fibrosis, the project aimed to isolate Extracellular Matrix (ECM) from human lung tissue using both oneday and three-day decellularization protocols to assess their impact on tissue integrity and future biomedical applications. • Executed precise decellularization processes and induced hypertension in porcine heart tissues, followed by comprehensive biochemical characterization of the ECM to evaluate its potential for drug testing, recellularization, and scaffold preparation. • Conducted a range of biochemical assays and mechanical testing, quantifying DNA, collagen, and glycosaminoglycans to compare the structural and biochemical properties of ECM isolated through short-term and extended decellularization protocols. • Generated detailed lab reports, critically analyzing the biomechanical and biochemical properties of ECM to refine decellularization techniques and enhance the reproducibility of results in future tissue engineering and fibrosis research. • Demonstrated superior biochemical and structural outcomes with the three-day decellularization method, providing valuable insights for the optimization of ECM-based scaffolds in therapeutic applications, including drug screening and recellularization strategies.
